چکیده

The nitrogenase system in cell-free extracts of soybean nodule bacteroids was fractionated into 2 components by use of protamine sulfate or polypropylene glycol precipitation followed by chromatography on DEAE-cellulose. Iron and molybdenum were concentrated in 1 fraction and iron in the other. Combination of fractions resulted in a striking stimulation in activity relative to the activity of individual fractions. The effect of different proportions of the 2 fractions on specific activities was studied. The ratios of the rates of reduction of acetylene and N(2) by extracts or fractions of different purities were relatively constant. Extracts or fractions retained most of their nitrogenase activities when stored in liquid N(2).

Effect of Gamma Irradiation on Some Biochemical Parameters and Physico-chemical Properties of Oil in Soybean [Glycine max (L.) Merr.] .

The soybean (Glycine max l. merr) is one of oil seed plants from leguminous family. In this study, the effect of gamma radiation on chlorophyll, soluble and insoluble carbohydrate and composition oil seeds were investigated in soybean. Soybean variety J.K was irradiated with 0,100,200 and 300 gamma ray. The treatment and control seeds planed in with four replicates. The highest amount of total ...

Adaptation of Nodulated Soybean (Glycine max L. Merr.) to Growth in Rhizospheres Containing Nonambient pO(2).

Nodulated soybean (Glycine max L. Merr. cv White Eye inoculated with Bradyrhizobium japonicum strain CB 1809) plants were cultured in the absence of combined N from 8 to 28 days with their root systems maintained continuously in 1, 2.5, 5, 10, 20, 40, 60, or 80% O(2) (volume/volume) in N(2). Plant dry matter yield was unaffected by partial pressure of oxygen (pO(2)) and N(2) fixation showed a b...

Effects of gradual increases in o(2) concentration on nodule activity in soybean.

The objectives of this study were to determine whether attached nodules of soybean (Glycine max L. Merr.) could adjust to gradual increases in rhizosphere pO(2) without nitrogenase inhibition and to determine whether the nitrogenase activity of the nodules is limited by pO(2) under ambient conditions. A computer-controlled gas blending apparatus was used to produce linear increases (ramps) in p...

Nitrogen fixation of nodulation mutants of soybean as affected by nitrate.

It was previously reported that three soybean (Glycine max [L.] Merr.) nodulation mutants (NOD1-3, NOD2-4, and NOD3-7) were partially tolerant to nitrate when nitrate was supplied simultaneously with inoculation at the time of transplanting. The current study evaluated the effect of short-term nitrate treatment on nitrogenase activity (C(2)H(2) reduction per plant and per nodule weight) and on ...

Immunoaffinity purification and comparison of allantoinases from soybean root nodules and cotyledons.

Allantoinase (allantoin amidohydrolase, EC 3.5.2.5) catalyzes the conversion of allantoin to allantoic acid in the final step of ureide biogenesis. We have purified allantoinase more than 4000-fold by immunoaffinity chromatography from root nodules and cotyledons of soybean (Glycine max [L] Merr.). We characterized and compared properties of the enzyme from the two sources. Seed and nodule alla...
